Problem: Which over the counter bacterial mouthwash, Listerine or Scope, is more effective in killing germs from a toothbrush used twice daily for one week.
Hypothesis: If you store a toothbrush used twice daily for one week in a glass container filled with Listerine, then there will be a significant bacterial reduction as compared to a toothbrush used twice daily for one week stored in a glass container filled with Scope.
Materials:


  1. 2 new toothbrushes

  2. 1 bottle of original Listerine

  3. 1 bottle of original Scope

  4. 10 petrie dishes

  5. 1 note book to record data

  6. 2 glass containers

  7. 1 clean bath towel

  8. 1 tube of Crest Whitening toothpaste

  9. 1 sharpie


Procedures:


  1. Lay out clean bath towel on bathroom counter top.

  2. Record the last food/fluid intake and time of intake in the data notebook.

  3. Label one toothbrush as A (bottom teeth) and the other toothbrush as B (top teeth).

  4. Fill 1 glass container with 2 inches of original Listerine, and the other glass container with 2 inches of original Scope.

  5. Take toothbrush A and apply a thin layer of Crest Whitening toothpaste to the toothbrush head, no water.

  6. Take toothbrush A and brush bottom teeth vigorously for 1 minute at 10 P.M.

  7. After completion of brushing, swab petrie dish label Day 1A PRE.

  8. After completion of swabbing, store toothbrush A head down in the container with 2 inches of Listerine.

  9. Take toothbrush B and apply a thin layer of Crest Whitening toothpaste to the toothbrush head, no water.

  10. Take toothbrush B and brush top teeth vigorously for 1 minute at 10 P.M.

  11. After completion of brushing, swab petrie dish label Day 1B PRE.

  12. After completion of brushing, store toothbrush B head down in the container with 2 inches of Scope.

  13. Store the toothbrushes in their respective containers from 10 P.M. to 6 A.M. the next morning.

  14. At 6 A.M. take toothbrush A and swab petrie dish labeled Day 1A POST.

  15. At 6 A.M. take toothbrush B and swab petrie dish labeled Day 1B POST.

  16. After the swabbing complete dump the two glass containers filled with Listerine and Scope and thoroughly rinse them out with warm water.

  17. For days 2-5 repeat steps 2-13 daily.

Questions:


  1. What is the best way to quantify bacterial growth? You will need to culture the bacteria. You will need Petri dishes with the appropriate agar to provide a nutrient and growing medium for the bacteria. You will culture the bacteria and count bacteria colonies. I should be able to get the agar plates you need from Shawnee Mission West.
